"Ron Davis and I were studying plant transcription factors in 1990, but progress was slow and arduous because the gene expression tools were so primitive. We conceived of the DNA microarray in 1993 to speed up gene expression analysis by combining biology and high technology. We adapted Affymetrix VLSIPS technology to synthesize yeast microarrays, and measured gene expression in yeast cells by hybridizing fluorescent probes derived from mRNA. The image shows the very first microarray experiment, with colored locations on the microarray providing readouts of expressed yeast genes. We worked frantically to improve the technology, and generated the first quantitative DNA microarray data using robotically printed plant cDNAs amplified by PCR. Science magazine kindly published our work (M. Schena et al., "Quantitative monitoring of gene expression patterns with a complementary DNA microarray." Science, 270:467-70, 1995), and since then the technology has exploded."

Arrayit Corporation has utilized genomic expression profiling and advanced bioinformatics to decipher the molecular basis of Parkinson's Disease (PD). In a collaboration with The Parkinson's Institute (Sunnyvale, CA), we compared messenger RNA levels in PD patients and controls, and identified a subset of 30 cellular genes dramatically up- and down-regulated in a PD-specific manner. Bioinformatic analysis of the gene signature revealed that Parkinson's Disease is caused by a perturbation of the protein folding and unfolding pathway, consistent with the formation of Lewy Body protein aggregates in patients with advanced PD. Based on the 30 gene markers, we developed the PDx molecular screening test that shows 100% sensitivity and 100% specificity in identifying Parkinson's Disease. PDx correctly detects the PD expression signature in symptomatic patients as well as in pre-symptomatic individuals 25 years before the appearance of symptoms, providing an extensive window for therapeutic intervention in pre-symptomatic patients. We are currently seeking a pharma or diagnostics strategic partner to advance PDx to a 510(k) FDA submission.

Arrayit offers the market's first service dedicated to deciphering rare human diseases. Rare (orphan) diseases are conditions presenting in a small percentage of the population, typically from 1 in 1,000-200,000 persons. More than 3,100 rare diseases have been identified but in many cases the molecular details are poorly understood. We deploy our patented and proprietary microarray platform on a genomic scale to elucidate the molecular basis of rare diseases quickly and efficiently. Control and test samples of mRNA are compared to produce a gene expression signature for each disease, and the gene expression data are examined computationally to produce a "best-fit" biological pathway. The sensitivity and specificity of our microarray platform enables a deeper examination of the transcriptome than competing platforms and a more complete understanding of rare/orphan diseases. Drug metabolism is a cornerstone of modern research and the pharmaceutical and healthcare industries because this metabolic process determines the rate of uptake, conversion of prodrugs into active drugs, and the speed by which active drug compounds are modified into less active metabolites in the human body. Drug metabolism is carried out primarily by enzymes located within the smooth endoplasmic reticulum of liver cells, though all cells have the biochemical capacity to metabolize drugs to a certain extent. Phase I metabolic reactions typically involve cytochrome P450 enzymes, resulting in the conversion of drugs from lipophilic to more hydrophilic compounds via oxidation and reduction reactions. Phase II reactions often involve cellular transferases, which add substituents such as methyl, sulfur, amino acid and other groups to aid in drug detoxification by facilitating excretion. Because drug metabolism is carried out by a large family of gene products, the ability to quantitatively measure drug metabolism gene expression is central to understanding this xenobiotic process.
