ArrayIt® has developed an innovative solution to multi-well microarrays that includes glass substrate slides containing a hydrophobic mask for well creation and a submersible hybridization cassette that allows temperature-controlled reactions without sample evaporation. Twenty four microarrays in a 3 x 8 pattern can be printed, reacted, scanned and quantified on a single glass substrate slide, offering highly affordable sample multiplexing for research and diagnostics.

Product Description
Designed for up to 24 high or low biochemical reactions on a single microarray substrate or slide. The unit is air tight to prevent evaporation and water proof, therefore short and extended reactions can be run in a high or low temperature controlled water bath or oven. This 24 well submersible hybridization cassette fits one 24 well masked substrate for 24 separate microarray reactions using a single cassette. No cover slips required.

This submersible cassette is made from a durable, flat, precision-machined corrosion resistant alloy and high quality black rubber gasket to ensure proper alignment and no leaking from “well to well”.

Figure 2.  Gasket matches the hydrophobic mask on the slide to guarantee no cross contamination between microarrays during the reactions.  Mask also assures that the microarray will release from the matching gasket easily when it is time to remove the microarrays from the cassette. Each well is 6 mm x 7 mm, capable of densities of tens to hundreds of capture analytes.  Rounded corners assures good coverage of reaction volume across all spots on the microarray. Mask does not interfere with microarray scans.

Usage Recommendations

  • Do not attempt to over fill each well. Overfilling can result in cross contamination.
  • After the desired reaction volume has been put in the wells, do not invert the reaction cassette upside down or rock at more than a 25-degree angle.  This can cause the reaction volume to stick to the lid that seals the wells closed and pull the reaction volume away from the microarray.  If this happens, tap the bottom of the entire cassette firmly on a lab bench to move the sample back down to cover the microarrays.
  • To view the reaction volume inside each well, keep the topside up and the cassette level. Then raise the cassette high enough to view the sample through the glass substrate.
  • Keep your reaction cassette clean with diluted mild detergents and clean water.  Sonication can be used.  Autoclave compatible.
  • When the lid that seals each well closed is removed, the entire base can be submerged in a wash buffer.  Lid can also be removed while the cassette is submerged in a wash buffer.  This product compatible with most all common laboratory buffer and detergent systems.

Gasket Information
Good resistance to all typical buffer systems used in DNA, Protein and other types of microarrays. Additionally the gasket is resistant to oils, UV and aging. Temperature range of the gasket material is -20˚C to 110˚C. Resistance to compression set is good. It is resistant to tearing or chipping is very good. Resistance to Aliphatic HC solvents is good, Aromatic HC is fair, Oxygenated HC is fair, Lacquer Solvents is poor, and Resistance to diluted acids and bases is excellent.

Benefits of Our Multiplexing Platform
1. Reduced cost and labor by multiplexing. Microarrays multiplex, miniaturize and parallelize many assay types. Microarrays have the ability to multiplex and miniaturize to a much greater degree. Our spotted microarray platform can simultaneously capture data from single analytes to tens of thousands of capture analytes in a single binding reaction, on a simple microscope slide size piece of glass. Standard microarray scanners can be used to read the results, these instruments are all over the world.

2. Shortened time-to-results by favorable reaction kinetics, with smaller sample requirements. Every spot in the microarray is subjected to the same processing conditions for great precision and consistent data among technical replicates. Common bead array plate based protocols use 50-100 ul reactions, each well is under separate processing and detection conditions. Smaller spots, less analyte and less detection reagents make microarray analysis less expensive and more accurate than bead arrays.

3. Planar reaction kinetics give faster, more reproducible results. For our spotted microarrays, the capture analytes are covalently bound to the surface of the microarray. They cannot interact with each other in solution, as can capture analytes attached to beads in solution. The miniaturization of spotted microarrays provide a platform where the detection DNA and protein molecules in solution are in far excess concentration than the capture analytes bound to the microarray. Increased concentration means faster binding kinetics and increased sensitivity. A properly executed microarray will provide a more sensitive and reproducible results than bead arrays.

4. Focused, flexible mutiplexing in the range of 1 to 1,000 analytes meets the needs of a wide variety of applications. Our spotted microarray have the ability for a high degree of miniaturization, number of capture analytes and fast data collection times. Microarrays use serum, amplified DNA, small molecules, carbohydrates, oligosaccarides, hybridomas, phage and others as capture analytes.

5. Open architecture allows market leading companies to provide compatible complimentary products. Our spotted microarray platform is an open architecture with microscope slide glass being the default substrate size used in microarray detection instruments and microarray manufacturing instruments. Microarray detection is routinely done with fluorescent, colorimetric and chemiluminescent detection...providing a flexible and open operating environment. Both CCD and laser/PMT architectures are available based on the end users need for quantitation, cost and sensitivity.
